Origin and history

Vitamin E was discovered in 1922 by Herbert Evans and Katherine Bishop at the University of California, Berkeley, who identified a fat-soluble factor essential for reproduction in rats. The active compound was later isolated and named tocopherol, from the Greek *tokos* (childbirth) and *pherein* (to bear). The d-alpha form is the most biologically active and the form preferentially retained by the human body.

Industrially, natural d-alpha tocopherol is extracted and purified from vegetable oils, primarily soybean, sunflower, and rapeseed oils. The process involves vacuum distillation and molecular distillation to concentrate the tocopherol fraction. Synthetic dl-alpha-tocopherol is produced from isophytol and trimethylhydroquinone. The natural form (d-) has approximately twice the biological activity of the synthetic form (dl-) on a per-milligram basis.

Scientific overview

Vitamin E functions primarily as a lipid-soluble antioxidant, scavenging peroxyl radicals and breaking the chain reaction of lipid peroxidation in cell membranes. It is incorporated into circulating lipoproteins and delivered to tissues via the liver's alpha-tocopherol transfer protein (α-TTP). This protein preferentially binds RRR-α-tocopherol (the natural d-form), which is why the natural form has higher bioavailability and biological activity than the synthetic dl-form.

The d-alpha form is the most bioavailable and biologically active of the eight naturally occurring Vitamin E compounds (four tocopherols and four tocotrienols). In gummy formulations, the acetate ester (d-alpha-tocopheryl acetate) is commonly used because it is more stable during processing and is hydrolysed to active tocopherol in the gut. The succinate ester is also available but less common in gummies due to solubility considerations.

From a manufacturing perspective, Vitamin E is heat-stable and oil-soluble, making it compatible with the lipid phase of gummy formulations. It does not degrade significantly under standard gummy cooking temperatures (80–100°C). The cost-per-mg is low, making it an economical addition to multivitamin blends. Because it is fat-soluble, it requires proper emulsification to ensure uniform distribution in the gummy matrix.
